Bis Maleimide Coupling

Bis maleimide coupling is a chemical strategy that uses a molecule containing two maleimide groups to connect thiol-bearing compounds, forming defined covalent bridges between molecular partners. Under mild conditions, thiolate nucleophiles add across the electron-deficient carbon-carbon double bond of each maleimide through a Michael addition, producing stable thioether linkages and enabling controlled conjugation or crosslinking. This method is widely used in bioconjugation to attach peptides, proteins, or other thiol-functionalized molecules, and in polymer and materials chemistry to build networks or linkers. Its predictable reactivity supports the preparation of engineered biomolecules, functional materials, and chemically tailored macromolecular structures.
