Cell Accumulation Assay

Cell accumulation assays are analytical methods used to measure how much of a chemical compound enters and remains within cells, providing a direct view of cellular exposure. In a typical assay, cultured cells are exposed to a defined compound concentration for a controlled period, then separated from the surrounding medium, washed, lysed, and analyzed to quantify intracellular material using fluorescence, radiotracing, or chromatography-based detection. In chemistry and pharmaceutical research, these assays help characterize membrane permeability, transporter activity, uptake kinetics, and compound retention. The resulting data support comparisons of molecular structure, formulation, and cellular delivery while informing studies of toxicity and drug action.

An Assay to Measure the Accumulation of a Fluorescent Antibiotic Probe in Bacterial Cells

This video demonstrates an assay to measure the accumulation of a fluorescent antibiotic probe in bacterial cells. Exposure to an efflux pump inhibitor disrupts the transmembrane proton gradient in bacterial cells, upon which a fluorophore-conjugated antibiotic is added. Inhibition of efflux pumps due to the disrupted proton gradient causes accumulation of the antibiotic inside the bacteria. Bacterial cells are lysed, and the fluorescence intensity is measured to confirm antibiotic accumulation.

Detection of Post-Replicative Gaps Accumulation and Repair in Human Cells Using the DNA Fiber Assay

Here we describe two modifications of the DNA fiber assay to investigate single-stranded DNA gaps in replicating DNA after lesion induction. The S1 fiber assay enables the detection of post-replicative gaps using the ssDNA-specific S1 endonuclease, while the gap-filling assay allows visualization and quantification of gap repair.
