Composition Analysis

Composition analysis is the systematic determination of which substances are present in a sample and the amounts or proportions of each component, making it fundamental to chemical identification and quality assessment. Chemists separate mixtures or measure characteristic interactions with matter using techniques such as chromatography, spectroscopy, titration, and mass spectrometry, then compare signals or reaction outcomes with standards to identify and quantify constituents. These methods support purity testing, formulation development, environmental monitoring, materials characterization, and pharmaceutical analysis. Reliable composition data also helps researchers trace contaminants, verify chemical structure, control manufacturing processes, and understand how molecular constituents influence a material’s properties.

A method for the determination of fatty acid content and composition in microalgae based on mechanical cell disruption, solvent based lipid extraction, transesterification, and quantification and identification of fatty acids using gas chromatography is described. A tripentadecanoin internal standard is used to compensate for the possible losses during extraction and incomplete transesterification.

Lipid polyesters constitute the structural components of two cell wall modifications, the plant cuticle and suberin-containing diffusion barriers. In this video, we describe a method to depolymerize cutin from whole delipidated leaves. The method can be applied to investigating mutants compromised in either cutin or suberin biosynthesis.
