Mosquito Ovary Development

Mosquito ovary development is the coordinated growth and maturation of ovarian follicles that enables egg production, a process central to mosquito reproduction and population biology. After a blood meal, hormonal signals such as juvenile hormone and ecdysteroids regulate vitellogenin synthesis, secretion, and uptake by developing oocytes, while amino acids, lipids, and carbohydrates provide materials for yolk formation and cellular growth. Studying these biochemical pathways connects chemistry with insect physiology and can reveal molecular targets for disrupting egg maturation, reducing mosquito fertility, and developing strategies to control disease-vector populations.

Temporal and spatial gene expression analyses have a crucial role in functional genomics. Whole-mount hybridization in situ is useful for determining the localization of transcripts within tissues and subcellular compartments. Here we outline a hybridization in situ protocol with modifications for specific target tissues in mosquitoes.

How niches and stem cells form during development is an important question with practical implications. In the Drosophila ovary, germ line stem cells and their somatic niches form during larval development. This video demonstrates how to dissect, stain and mount female gonads from late third instar (LL3) Drosophila larvae.

This protocol describes the primary culture/co-culture of mouse ovarian tissue, using ovaries from neonatal mice and individual ovarian follicles from prepubertal mice. The culture techniques support development in a highly physiological manner, allowing investigation of the effect of extrinsic agents on the ovary, and of interactions between ovarian follicles.
