Fraction Metabolized

Fraction metabolized is the proportion of a drug dose cleared through metabolic pathways rather than excreted unchanged, making it an important pharmacokinetic measure in clinical drug development and therapy. It reflects the balance between enzymatic biotransformation, often by hepatic CYP450 or conjugation enzymes, and alternative routes such as renal elimination of unchanged drug; changes in enzyme activity can therefore alter drug exposure. Estimating fraction metabolized helps predict the effects of enzyme inhibitors or inducers, assess patient-to-patient variability, and anticipate how liver or kidney dysfunction may influence dosing, safety, and drug interactions.

Mitochondria play central roles in the regulation of metabolism and homeostasis. Subtle changes in mitochondrial metabolism that affect organismal physiology could be difficult to detect in whole organism metabolomics studies. Here we describe an isolation method that enhances the detection of subtle metabolic shifts in Drosophila melanogaster.

Here we describe a robust method for the fractionation of plant plasma membranes into detergent resistant and detergent soluble membranes based on a mixture of unlabeled and in vivo fully 15N labeled Arabidopsis thaliana cell cultures. The procedure is applied for comparative proteomic studies to understand signaling processes.

Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.
