Activin Nodal Signaling Inhibitors

Activin Nodal signaling inhibitors are molecules that suppress transforming growth factor-beta (TGF-β) pathway signals controlling cell fate, tissue patterning, and embryonic development. They act by blocking ligand-receptor interactions, inhibiting activin or Nodal receptor kinase activity, or preventing phosphorylation and nuclear activity of downstream SMAD2 and SMAD3 transcription factors. In developmental biology, these inhibitors help dissect how concentration, timing, and spatial distribution of Activin-Nodal signals regulate germ-layer formation, left-right patterning, and stem cell differentiation. They are also used to control pluripotent cell states and improve the reproducibility of organoid and in vitro differentiation models.

In this video, we describe a method to identify the small-molecule kinase inhibitors that modulate the apoptosis of self-reactive CD4+CD8+ double-positive immature thymocytes. Apoptosis is induced in the double-positive thymocytes by activating them with anti-CD3- and anti-CD28-coated magnetic beads; this is followed by a small-molecule inhibitor treatment and flow cytometry analysis to detect if the inhibitors modulate the apoptotic marker expression.
