Pollen Grain Examination

Pollen grain examination is the microscopic analysis of pollen structure, development, viability, and function, providing insight into the male reproductive phase of flowering plants. Researchers typically collect pollen at defined developmental stages, prepare and stain grains for light or electron microscopy, and assess features such as wall architecture, apertures, cellular organization, and germination capacity. In developmental biology, these observations help track microsporogenesis, pollen maturation, and the formation of the male gametophyte, while viability and germination assays reveal how successfully pollen can support fertilization. The approach also supports studies of reproductive abnormalities, plant fertility, taxonomy, and environmental effects on development.

A High-Resolution, Single-Grain, In Vivo Pollen Hydration Bioassay for Arabidopsis thaliana

An improved method to measure pollen hydration profiles in Arabidopsis thaliana is described here. The new method offers higher resolution, is noninvasive, and is highly reproducible. The protocol represents a new tool for a finer dissection of the processes that regulate the early stages of pollination.

In this work, we describe the use of the atom-probe tomography technique for studying the grain boundaries of the absorber layer in a CIGS solar cell. A novel approach to prepare the atom probe tips containing the desired grain boundary with a known structure is also presented here.

Characterization of Ultra-fine Grained and Nanocrystalline Materials Using Transmission Kikuchi Diffraction

This paper provides a detailed method to characterize the microstructure of ultra-fine grained and nanocrystalline materials using a scanning electron microscope equipped with a standard electron backscatter diffraction system. Metal alloys and minerals presenting refined microstructures are analyzed using this technique, showing the diversity of its possible applications.
