Thp-1 Monocytes

THP-1 monocytes are an immortalized human monocytic cell line derived from acute monocytic leukemia and widely used to study monocyte biology in vitro. They grow in suspension and can be induced to differentiate into adherent macrophage-like cells, commonly with phorbol 12-myristate 13-acetate (PMA), which activates protein kinase C-dependent signaling and alters gene expression, morphology, and immune function. In developmental biology, this transition provides a controllable model for examining myeloid differentiation, cell maturation, and responses to environmental cues, although the resulting cells do not fully reproduce primary human monocytes or macrophages. THP-1 systems support mechanistic studies of lineage development and immune regulation, as well as standardized testing of cellular pathways and stimuli.

This protocol serves as a detailed guide to reproducibly differentiate and polarize THP-1 monocyte-like cells into M2-like macrophages within 14 days. This model is the basis to investigate the anti-inflammatory effects of TAM within the tumor microenvironment.

This protocol is based on LPS and ATP-induced death of PMA-differentiated THP-1 macrophages. We use flow cytometry to analyze Annexin V and 7-AAD double staining to detect cell death, using the whole cell and employing scanning electron microscopy to observe the cell membrane morphology.

We have developed an in vitro malaria-HIV-1 co-infection model to study the impact of Plasmodium falciparum on the HIV-1 replicative cycle in human primary monocyte-derived macrophages. This versatile system can easily be adapted to other primary cell types susceptible to HIV-1 infection.
