Revolving Rollers

Revolving rollers are cylindrical engineering components that rotate about fixed or moving axes to support, transport, guide, or shape materials while reducing sliding friction. Their operation relies on rolling contact: bearings or shafts constrain the rollers, and applied force, gravity, or an external drive produces rotation that transfers loads and motion through the contact surface. Revolving rollers are used in conveyor systems, material-handling equipment, manufacturing lines, and forming processes, where their diameter, spacing, surface texture, alignment, and rotational speed affect stability, traction, throughput, and wear. Studying these variables helps engineers improve efficiency, control material flow, and design reliable mechanical systems.

A Modified Roller Tube Method for the Long-Term Culture of Rodent Brain Slices

2025

This video demonstrates a modified roller tube method for the long-term culturing of brain slices from a rodent model. First, the brain slices are attached to coverslips. Then, flat-sided roller tubes with a hole on the flat side are taken, and the coverslips are positioned on the hole using adhesive discs. Finally, a nutrient medium and a carbon dioxide and air mixture are added to the tube, and the tubes are incubated with a rolling motion.

Presented here is a modified roller tube method for culturing and intermittent high-resolution imaging of rodent brain slices over many weeks with precise repositioning on photoetched coverslips. Neuronal viability and slice morphology are well maintained. Applications of this fully enclosed system using viruses for cell-type specific expression are provided.

Understanding the neural substrates of behavior requires brain circuit ensemble recording. Because of its genetic tractability, the mouse offers a model for circuit dissection and disease mimicry. Here, a method of designing and fabricating miniaturized probes is described that is suitable for targeting deep brain structure in the mouse.

Targeted Neuronal Ablation Using Two-Photon Microscopy in a Zebrafish Larava

Source: Muto, A., et al. Ablation of a Neuronal Population Using a Two-photon Laser and Its Assessment Using Calcium Imaging and Behavioral Recording in Zebrafish Larvae. J. Vis. Exp. (2018) In this video, a two-photon laser ablation procedure is demonstrated, enabling the selective destruction of fluorescent neurons in agarose-immobilized zebrafish larvae. High-resolution imaging before and after the procedure confirms efficient neuronal damage, as observed by the absence of fluorescence.
