Digital Twin Analysis

Digital twin analysis is the use of a dynamically updated virtual representation of a physical object, process, or environment to understand its current state and anticipate changes. It works by integrating observations from sensors, monitoring systems, or other data sources with computational models; the digital counterpart is then compared with the physical system and refined as new data arrive. In environmental research, this approach can represent ecosystems, watersheds, cities, or industrial sites, allowing researchers to test scenarios, identify anomalies, assess resource use, and evaluate management strategies before implementing them in the real world. Its value lies in linking continuous observation with evidence-based prediction and decision-making.

Here, we present a protocol for a mosaic labeling technique that permits the visualization of neurons derived from a common progenitor cell in two distinct colors. A method is described for efficient purification of twin-Strep-tagged fusion proteins and their specific complexes on modified streptavidin (Strep-Tactin) resin covalently cross-linked with Bis(sulfosuccinimidyl) suberate (BS3). The method has the advantages of fast speed, good target protein recovery and high purity, and is compatible with subsequent analysis by mass spectrometry.

Automated Sholl Analysis of Digitized Neuronal Morphology at Multiple Scales

We have developed a computer program to analyze neuronal morphology. In combination with two existing open source analysis tools, our program performs Sholl analysis and determines the number of neurites, branch points, and neurite tips. The analyses are performed so that local changes in neurite morphology can be observed.
