Break-induced Replication

Break-induced replication (BIR) is a DNA repair and replication process that restarts DNA synthesis from a single-ended double-strand break, helping cells recover from collapsed replication forks and repair damaged chromosomes. It begins when a broken DNA end invades a homologous sequence, forming a displacement loop that recruits replication machinery; the nascent strand then undergoes extensive, often conservative DNA synthesis, sometimes with template switching. In genetics, BIR explains how cells maintain chromosome continuity while also generating mutations, loss of heterozygosity, copy-number changes, and rearrangements. Studying this pathway clarifies genome instability mechanisms relevant to inherited disease and cancer biology.

Inducing a Site Specific Replication Blockage in E. coli Using a Fluorescent Repressor Operator System

We describe here a system utilizing a site-specific, reversible in vivo protein block to stall and collapse replication forks in Escherichia coli. The establishment of the replication block is evaluated by fluorescence microscopy and neutral-neutral 2-dimensional agarose gel electrophoresis is used to visualize replication intermediates.

DNA replication involves the separation of the two strands of the double helix, with each strand serving as a template from which the new complementary strand is copied. After replication, each double-stranded DNA includes one parental or “old” strand and one “new” strand. This is known as semiconservative replication. The resulting DNA molecules have the same sequence and are divided equally into the two daughter cells. Replication in Prokaryotes DNA replication uses a large number of...

The macroeconomic consumption function illustrates the relationship between aggregate consumption (C) and national income (Y). It is typically expressed as:C = a + bYwhere a denotes autonomous consumption—expenditures that occur regardless of income—and b, the marginal propensity to consume (MPC), indicates the fraction of additional income that is spent. The break-even point occurs where:C = YSolving for income yields the break-even level of national income:At this level, the entire output...
