Bead Based Multiplex Assay

A bead-based multiplex assay is a laboratory method that measures multiple analytes simultaneously by using distinct populations of microscopic beads, making it valuable for profiling immune responses and infection-related biomarkers. Each bead population carries a specific capture antibody or probe and an identifying fluorescent code; target molecules bind during incubation, then labeled detection reagents generate signals proportional to analyte abundance. Flow-based or fluorescence-based instruments distinguish bead identities and quantify several cytokines, antibodies, antigens, or nucleic acid targets from a single sample. This approach conserves specimen volume, increases analytical efficiency, and supports immune monitoring, pathogen characterization, diagnostic research, and evaluation of vaccine or therapeutic responses.

This video demonstrates a bead-based multiplex immunoassay technique to simultaneously detect antibodies for multiple environmental pathogens in human saliva. Fluorescent beads were coupled to antigens from different pathogens, which were captured using antibodies in the human saliva sample. Upon labeling the antibodies using a fluorescent reporter, the beads were analyzed using flow cytometry to assess the presence of different pathogen-specific antibodies in the saliva.
