Hemolymph Perfusion

Hemolymph perfusion is the controlled movement or exchange of hemolymph, the circulatory fluid of arthropods and other invertebrates, through the body or isolated tissues. In an open circulatory system, a dorsal vessel and body movements drive hemolymph through the hemocoel, where it bathes organs and transports hemocytes, nutrients, signaling molecules, and immune factors. Laboratory perfusion can maintain tissues, enable controlled exposure to pathogens or immune stimuli, and support collection of hemolymph for cellular and biochemical analysis. In immunology and infection research, these approaches help characterize hemocyte activity, antimicrobial defenses, pathogen dissemination, and host responses under defined experimental conditions.

Cuticle Disruption: A Method to Collect Hemolymph from Drosophila Larvae

2023

The Drosophila hemolymph serves as both blood and interstitial fluid, circulating throughout the body during the larva and adult stages. This video describes a protocol to collect larval hemolymph in order to quantify the circulating cells.

Insulin Injection and Hemolymph Extraction to Measure Insulin Sensitivity in Adult Drosophila melanogaster

Conserved insulin signaling pathways found in the fruit fly Drosophila melanogaster make this organism a potential tool for modeling metabolic disorders including type II diabetes. To this end, it is critical to establish physiological assays to effectively measure systemic insulin action in peripheral glucose disposal in the adult fly.

Here are methods to quantify nutrients in pollen before and after its conversion to beebread by two subspecies of honeybees. We describe techniques to measure beebread consumption and resulting protein titers in both subspecies.

Antegrade Perfusion Model: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

2025

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.

Langendorff Retrograde Heart Perfusion: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

2025

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.
