Lps Treatment Protocol

An LPS treatment protocol is a standardized method for exposing immune cells or experimental organisms to lipopolysaccharide, a bacterial outer-membrane component used to model innate immune activation. LPS binds the CD14–MD-2–TLR4 receptor complex, triggering signaling pathways such as NF-κB and leading to the production of inflammatory mediators. Researchers adjust LPS concentration, exposure time, and experimental controls to examine cytokine responses, immune-cell activation, and inflammation. In immunology and infection research, this approach helps characterize host–pathogen interactions, compare inflammatory phenotypes, and evaluate candidate anti-inflammatory or immunomodulatory treatments.

Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.

This video demonstrates an in vitro technique to study the impact of antimicrobial treatment on Pseudomonas aeruginosa (Pa) aggregates. Fluorescently-labeled Pa cells are grown as aggregates, mimicking the growth pattern during infection. Upon challenging the aggregates with a sublethal concentration of the antibiotic colistin, the real-time assessment of the bacteria-antibiotic interaction is performed via fluorescence microscopy.
