Modified Ldh Assay

The modified LDH assay is a colorimetric method for measuring cell damage or cytotoxicity by detecting lactate dehydrogenase released from cells with compromised plasma membranes. In immunology and infection studies, released LDH catalyzes reactions that generate a measurable color signal, whose intensity reflects the amount of membrane disruption in the sample. Modifying the assay format can improve its suitability for particular cell types, infection models, or experimental controls while reducing the need for radioactive labels. Researchers use the resulting measurements to evaluate pathogen-associated injury, immune-cell cytotoxicity, host-cell viability, and the effects of antimicrobial or immunomodulatory treatments.

Analysis of Trunk Neural Crest Cell Migration using a Modified Zigmond Chamber Assay

An approach to analyze the migration of explanted cells (trunk neural crest cells) is described. This method is inexpensive, gentle, and capable of distinguishing chemotaxis from both chemokinesis and other influences on migratory polarity such as those derived from cell-cell interactions within the primary trunk neural crest cell culture.

Modified Yeast-One Hybrid Assay to Detect Heteromeric Protein Complex-DNA Interactions

In this video, we demonstrate the modified yeast one-hybrid assay, which detects multiple proteins interacting with target DNA. The transformed yeast cells contain the target DNA sequence upstream of the reporter gene. Additionally, they express transcription factor proteins. When the proteins form complexes and bind to the target DNA, the reporter gene is expressed and can be detected enzymatically.

In Vivo Alkaline Comet Assay and Enzyme-modified Alkaline Comet Assay for Measuring DNA Strand Breaks and Oxidative DNA Damage in Rat Liver

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.

An accurate method for the assessment of cell death is described. The protocol improves upon conventional Annexin V/ propidium iodide (PI) protocols, which display up to 40% false- positive events in cell lines and primary cells from a broad range of animal models.

This video demonstrates the lactate dehydrogenase assay to detect inflammasome-mediated macrophage death. The macrophages are exposed to lipopolysaccharide for priming, followed by exposure to nigericin to cause inflammasome-induced cell death, which is determined by the lactate dehydrogenase assay.
