Photoconversion-based Tracking

Photoconversion-based tracking is an imaging approach that uses light to mark selected cells or regions with photoconvertible fluorescent labels, enabling their movement and fate to be followed over time. Focused illumination irreversibly shifts the label’s fluorescence from one spectral state to another, creating a time-stamped population that can be identified during subsequent microscopy or flow-based analysis. In immunology and infection research, this method reveals how immune cells migrate between tissues, interact with infected sites, and contribute to local or systemic responses. It can clarify cell trafficking, retention, and dispersal in complex biological environments while preserving spatial information.

Photoconversion-Based Leukocyte Tracking in Bacterial-Infected Transgenic Zebrafish

2025

This video demonstrates the tracking of leukocyte recruitment using transgenic zebrafish larvae expressing photoconvertible green fluorescent protein. Pre-injecting red fluorescent dye-labeled bacteria into the otic vesicle and inducing photoconversion enables observation of leukocytes' migration, phagocytosis, and dispersal throughout the body, providing valuable insights into immune response dynamics.

We established the photoconvertible PSmOrange system as a powerful, straight-forward and cost inexpensive tool for in vivo cell tracking in GFP transgenic backgrounds. This protocol describes its application in the zebrafish model system.

The Photoconversion Technique for Exploring Inflammatory Cell Dynamics in Insect Pupae

The video demonstrates the use of photoconversion to study inflammatory cell dynamics in Drosophila pupae. Green hemocytes are drawn to the wounded epithelium, followed by distant migration. Photoconversion shifts certain hemocytes from green to red, facilitating their movement tracking.
