Cell Integrity

Cell integrity is the preservation of a cell’s structure, boundaries, and coordinated functions, a fundamental requirement for healthy tissues and organs. It depends on an intact plasma membrane, cytoskeleton, organelles, and regulatory systems that maintain ion gradients, control transport, support energy production, and respond to stress. Physical injury, toxins, infection, inflammation, or metabolic imbalance can disrupt these mechanisms, causing membrane leakage, impaired signaling, organelle dysfunction, or cell death. In medicine, assessing cell integrity helps researchers and clinicians evaluate tissue damage, drug toxicity, disease progression, and treatment responses, supporting safer therapies and more accurate models of human health.

This protocol describes a detailed method for efficient generation of integration-free iPSCs from human adult peripheral blood cells. With the use of four oriP/EBNA-based episomal vectors to express the reprogramming factors, KLF4, MYC, BCL-XL, or OCT4 and SOX2, thousands of iPSC colonies can be obtained from 1 mL of peripheral blood.

Integration by Parts: Indefinite Integrals

Integration by parts is a fundamental technique in calculus for evaluating integrals involving the product of two functions. It is particularly useful when direct integration is not feasible. The method is based on the product rule for differentiation, which states that the derivative of a product equals the derivative of the first function times the second, plus the first function times the derivative of the second. By integrating this identity and rearranging terms, the integration by parts...

Integration by Parts: Definite Integrals

Definite integrals involving the product of two functions over a fixed interval can be evaluated using integration by parts. This method rewrites the integral as the difference of a product evaluated at the endpoints and a remaining definite integral that is often simpler to compute.A representative example is the definite integral of the inverse tangent function. Since there is no direct integration formula for arctan ⁡x, the integrand is rewritten as a product of arctan⁡ x and the constant...

Induced pluripotent stem cells (iPSCs) represent a source of patient-specific tissues for clinical applications and basic research. Here, we present a detailed protocol to reprogram human peripheral blood mononuclear cells (PBMNCs) obtained from frozen buffy coats into viral-free iPSCs using non-integrating episomal plasmids.
