Human Ku80 Staining

Human Ku80 staining is a laboratory method for detecting the Ku80 protein in human cells or tissue, providing insight into DNA double-strand break repair and cellular responses to genomic damage. The technique uses antibodies that bind specifically to Ku80, a component of the Ku heterodimer that recognizes broken DNA ends and helps recruit DNA-dependent protein kinase during non-homologous end joining; labeled secondary antibodies then reveal its abundance and subcellular localization by fluorescence or microscopy. In medicine, Ku80 staining supports studies of tumor biology, DNA repair proficiency, treatment responses, and disease-associated changes in nuclear protein expression.

Staining Protocols for Human Pancreatic Islets

This video demonstrates procedures for characterization of human pancreatic islets using hematoxylin and eosin (H&E) and immunohistochemistry (IHC). Pancreatic sections from head, body, and tail regions are stained by both H&E and IHC to determine islet endocrine composition (insulin, glucagon, and pancreatic polypeptide), cell replication (Ki67), and inflammatory infiltrates (H&E, CD3). The uncinate region is localized using IHC for pancreatic polypeptide.

Production of Tissue Microarrays, Immunohistochemistry Staining and Digitalization Within the Human Protein Atlas

Tissue microarrays allows for an efficient method to gain concurrent information from a multitude of tissues. Representative parts of tissues are assembled into a single paraffin block. Sections from the block are used for immunohistochemistry and analysis of protein expression patterns. Digital scanning generates corresponding images for distribution of data.

This video demonstrates the immunohistochemical staining for S-100 protein in rectal suction biopsy sections. Antigen-retrieved sections are blocked and treated with primary, followed by secondary antibodies against S-100, a marker of Hirschsprung's disease. Chromogenic substrate is added to stain the targeted cells. Samples are dehydrated and mounted before visualizing under the microscope.

DNA-FISH Metaphase Spreads to Distinguish Extrachromosomal DNA from Homogeneously Staining Regions in Human Cancer Cell Lines

We report a DNA-FISH metaphase spread protocol that visually detects locus copy number and location within the genome. This approach enables single-cell resolution of amplification states, specifically in cancer cell lines containing extrachromosomal DNA and homogeneously staining regions.
