Mono-iodoacetate Injection

Mono-iodoacetate injection is a chemical method used to induce localized joint damage and pain, most commonly for studying osteoarthritis in laboratory animals. After intra-articular administration, mono-iodoacetate inhibits glyceraldehyde-3-phosphate dehydrogenase in chondrocytes, disrupting glycolysis and cellular energy production; subsequent chondrocyte death contributes to cartilage degeneration, inflammation, and pain-related behavior. This model reproduces important features of degenerative joint disease and provides a controlled system for investigating disease mechanisms, nociceptive pathways, structural changes, and functional impairment. Researchers also use it to evaluate analgesic, anti-inflammatory, and potential disease-modifying treatments before clinical investigation.

Pseudomonas aeruginosa produces the rhamnolipid biosurfactants. Thin-layer chromatography detects and determines the proportion of mono- and di-rhamnolipids produced by each strain. Quantification of total rhamnolipids involves assessing rhamnose equivalents present in these biosurfactants extracted from the culture supernatants using the orcinol method.

MicroRNAs play crucial roles in the brain and are potential targets for modeling neuro-degeneration. However, perturbing miRNA levels is challenging due to the short length of miRNA and inaccessibility of the brain tissue. This video presents a method for antagomir design and brain specific delivery using a neuropeptide in mice.

Lymph nodes are the immunological tissues that orchestrate immune response and are a critical target for vaccines. Biomaterials have been employed to better target lymph nodes and to control delivery of antigens or adjuvants. This paper describes a technique combining these ideas to inject biocompatible polymer particles into lymph nodes.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.
