Partial Carotid Ligation

Partial carotid ligation is a surgical technique that reduces blood flow through the carotid artery to create a controlled model of vascular disease. In commonly used animal models, selected carotid branches are ligated while one branch remains open, producing low, disturbed, and oscillatory shear stress along the arterial wall. This altered hemodynamic environment promotes endothelial dysfunction, inflammation, and atherosclerotic plaque formation, allowing researchers to study how abnormal blood flow initiates and accelerates vascular pathology. The model supports investigation of atherosclerosis, vascular remodeling, inflammatory signaling, and potential therapies for cardiovascular disease in a reproducible experimental setting.

This describes a partial carotid ligation surgery, which causes disturbed flow conditions and subsequent atherosclerosis development (in two weeks) with intraplaque neo-vascularization (in four weeks) in the mouse common carotid artery. We also describe a novel method of RNA isolation from the carotid intima, providing high purity endothelial RNA.

This protocol describes a robot-assisted partial splenectomy for a benign splenic tumor. The robotic approach optimizes spleen preservation by enhancing surgical precision, minimizing blood loss, and improving postoperative outcomes. Key surgical steps include meticulous splenic hilum dissection, selective vascular ligation, intraoperative ultrasonography, and controlled parenchymal transection.
