Percoll Gradient Purification

Percoll gradient purification is a density-based centrifugation method that separates cells, organelles, or particles according to their buoyant density, making it valuable for isolating biologically relevant populations in medical research. The sample is layered onto a gradient of Percoll, colloidal silica particles coated with polyvinylpyrrolidone, and centrifuged so components migrate to positions where their density matches the surrounding medium. Because Percoll gradients can be formulated under isotonic conditions, they help maintain cell structure and viability during separation. Applications include preparing purified leukocyte, stem cell, sperm, and organelle fractions for diagnostics, transplantation studies, immunology, and other biomedical investigations.
