Porcine Tibial Allotransplantation Model

The Porcine Tibial Allotransplantation Model is an in vivo research system that uses donor and recipient pigs to study the transplantation of tibial bone between genetically distinct animals. During the procedure, a donor tibial segment is implanted into a recipient, allowing researchers to examine graft incorporation, revascularization, bone remodeling, and immune recognition under physiologically relevant conditions. This model supports investigation of rejection mechanisms, immunosuppressive strategies, graft preservation, and surgical reconstruction. Because porcine bone size, structure, and healing responses resemble important aspects of human orthopedics, the model helps evaluate treatments and transplantation protocols before clinical application.

Currently any kind of vascularized composite allotransplantation depends on long-term-immunosuppression, difficult to support for non-life-critical indications. We present a new porcine tibial VCA model that can be used to study bone VCA and demonstrate the use of surgical angiogenesis to maintain bone viability without the need of long-term immune-modulation.

Source: Brakkee, E. M., et al. Surgery and Behavioral Testing in the Tibial Neuroma Transposition Model in Rats. J. Vis. Exp. (2023).This video demonstrates a procedure to generate a rat tibial neuroma transposition (TNT) model. After exposing the tibial nerve in the hind leg of an anesthetized rat, transect the nerve. Next, create a subcutaneous tunnel, thread the severed nerve through the tunnel, and suture it in place. In the absence of a target for reinnervation, disorganized axonal...

This protocol describes the tibial neuroma transposition model, which entails a lesion of the tibial nerve with subsequent transposition of the proximal nerve end toward a subcutaneous pretibial or lateral position. Behavioral testing of neuroma pain and plantar hyperalgesia is quantified using Von Frey monofilaments.

This protocol provides detailed guidance for the initial and continued generational allotransplantation of Drosophila tumors into the abdomen of adult hosts for studying various aspects of neoplasia. Using an autoinjector apparatus, researchers can achieve improved efficiency and tumor yields compared to those achieved by traditional, manual methods.
