Tissue Culture Methods

Tissue culture methods are laboratory techniques for maintaining, growing, and studying cells or tissue fragments under controlled conditions, providing a model of human biology outside the body. Researchers place cells or tissue explants in sterile nutrient media and regulate factors such as temperature, pH, oxygen, and growth signals to support survival, proliferation, or differentiation. In medicine, these methods help investigate disease mechanisms, evaluate drug responses and toxicity, and examine tissue repair or regeneration. They also support the development of cell-based therapies and personalized models, although results must be interpreted alongside the complexity of living tissues and patients.

This video introduces histotypic tissue culture, where the growth and propagation of one cell line is done in an engineered three-dimensional matrix to reach high cell density. Whole Organ Tissue Culture

Whole organs can be cultured ex vivo using specialized bioreactors with the goal of repairing or replacing entire organs. This method uses a donor organ that is stripped of all cells, leaving behind the three-dimensional structure, which is then repopulated with new cells. Here, we present an organotypic culturing protocol to grow embryonic chicken organs in vitro. Using this method, the development of embryonic chicken tissue can be studied, while maintaining a high degree of control over the culture environment.
