Bone Tissue Sections

Bone tissue sections are thin, prepared slices of bone used to examine its microscopic structure and interactions with surrounding tissues. Because mineralized bone is hard, samples are typically fixed, decalcified or embedded in a supportive medium, cut with specialized equipment, and stained to reveal cells, matrix, blood vessels, and neural elements. In neuroscience, these sections help researchers study bone–nerve interfaces, cranial and spinal tissues, sensory innervation, and changes associated with injury or disease. Microscopic analysis can connect tissue architecture with neural function and support investigations of regeneration, pain, and neuro-orthopedic disorders.

In this video, we describe a technique of hematoxylin and eosin staining of paraffin-embedded tissue sections to study their histology. The procedure includes deparaffinization of the tissue followed by staining the cells in the tissue. Upon staining, the nucleus appears distinctly blue, and the proteins in the cytoplasm display pink coloration.
