Axin2-lacz Reporter

The Axin2-lacZ reporter is a genetically engineered system that marks cells expressing Axin2, enabling visualization of canonical Wnt signaling in tissues such as the nervous system. In this reporter, regulatory elements from the Axin2 gene drive lacZ expression, producing β-galactosidase in response to Wnt/β-catenin pathway activity; staining therefore reveals the location and relative distribution of pathway-responsive cells. In neuroscience, Axin2-lacZ helps map Wnt signaling during brain development, identify neural progenitor and stem-cell populations, and examine pathway changes after injury or disease. This approach links molecular signaling patterns with neural cell specification, tissue organization, and regeneration.

2025

This video demonstrates the measurement of retinoic acid (RA) secretion from neurosphere cells using RA reporter cells. It outlines the steps involved in co-culturing neurosphere cells with RA reporter cells and performing a colorimetric RA assay to measure RA levels.

Employing Reporter Cells in a Co-Culture to Study Retinoic Acid Production by Mouse Embryonic Cells

This video demonstrates a technique for assessing retinoic acid activity in mouse embryos. Upon isolating cells from the mouse embryos, the cells are co-cultured with retinoic acid (RA) reporter cells. Embryonic cells produce RA, which induces beta-galactosidase expression in reporter cells, indicating RA activity in the embryonic cells.

Flow cytometry is a powerful tool allowing for the isolation and study of specific cell populations. This protocol describes steps for isolating LacZ-expressing cells from cochlear tissues from neonatal transgenic mice. Dissociated cochlear cells were labeled using fluorescent-conjugated substrates of β-galactosidase prior to separation via flow cytometry.

Data Reporting and Recording

Reporting and recording are crucial in data documentation. The timely, thorough, and accurate documentation of facts is essential when recording patient data. Failure to record findings during an assessment or interpretation of a problem will result in loss of information and make the patient document unreliable. The reader is left with general impressions if the information is not specific. A recording is documenting data of the individual's health information in a traceable, secure, and...
