Brain Mitochondria Imaging

Brain mitochondria imaging is the visualization and measurement of mitochondria within neurons and other brain cells, enabling researchers to study how these organelles support cellular energy and respond to stress. The approach uses fluorescence-based dyes, genetically encoded reporters, or other microscopy-compatible markers to reveal mitochondrial location, morphology, movement, membrane potential, or respiratory activity in living or fixed tissue. In neuroscience, these measurements can connect mitochondrial dynamics and dysfunction with synaptic activity, neurodevelopment, aging, and neurological disease. Quantitative imaging also helps evaluate how genetic changes, metabolic conditions, or candidate treatments alter mitochondrial health while linking subcellular processes to brain-cell function.

Here, we describe how three dimensional (3D) reconstruction analysis from the serial block-face scanning electron microscopy (SBFSEM) can be used to gain insights on the morphological and volumetric analysis of this critical energy generating organelle.

A spectrofluorometric protocol for the measurement of the mitochondrial permeability transition pore opening in isolated mouse heart mitochondria is presented here. The assay involves the simultaneous measurement of mitochondria Ca2+ handling, mitochondrial membrane potential and mitochondrial volume. The procedure for obtaining high-quality and functional heart mitochondria is also described.
