Brain Section Preparation

Brain section preparation is a set of laboratory methods used to preserve, support, and thinly slice brain tissue for microscopic examination, making cellular structure and spatial organization accessible to researchers. The process typically involves fixation to stabilize tissue, followed by cryoprotection and freezing or dehydration and embedding, then sectioning with a cryostat or microtome; collected sections may be stained or labeled for imaging. In neuroscience, prepared sections enable analysis of neurons, glial cells, brain regions, and pathological changes, supporting studies of development, neural circuits, injury, and disease while providing the anatomical context needed to interpret molecular and behavioral findings.

We describe a protocol for transcardiac perfusion of mice, removal and sectioning of the brain, as well as immunoperoxidase staining, resin embedding, and ultrathin sectioning of the brain sections. Upon completion of these procedures, the immunostained material is ready for examination with transmission electron microscopy.
