Glial Cell Engulfment

Glial cell engulfment is the process by which nervous-system glia internalize and degrade cells, synapses, myelin, or other extracellular material, helping maintain neural tissue. In this form of cellular phagocytosis, glial receptors recognize exposed molecular signals or injury-associated cues, trigger cytoskeletal remodeling and membrane extension around the target, and deliver the enclosed material to lysosomes for degradation. Microglia are major engulfing cells, while astrocytes can also remove synaptic elements and cellular debris. Studying this process clarifies synaptic pruning, development, injury responses, and neurodegenerative disease, and may identify mechanisms that can be targeted to preserve neural connections or improve tissue repair.

Overview Glial cells are one of the two main types of cells in the nervous system. Glia cells comprise astrocytes, oligodendrocytes, microglia, and ependymal cells in the central nervous system, and satellite and Schwann cells in the peripheral nervous system. These cells do not communicate via electrical signals like neurons do, but they contribute to virtually every other aspect of nervous system function. In humans, the number of glial cells is roughly equal to the number of neurons in the...

Microglia are the resident immune cells of the central nervous system (CNS) with a high capacity to phagocytose or engulf material in their extracellular environment. Here, a broadly applicable, reliable, and highly quantitative assay for visualizing and measuring microglia-mediated engulfment of synaptic components is described.

Isolating Enteric Glial Cells From the Submucosa and Lamina Propria of a Mouse

This video demonstrates a method for isolating enteric glial cells from the submucosa and lamina propria of the mouse intestine. It outlines the steps involved in extracting submucosa and lamina propria tissue, isolating the enteric glial cells from them, and culturing the cells on a coated well-plate to establish an enteric glial cell culture.

Immunofluorescence Staining of Drosophila Brains for the Single-Cell Imaging of Glial Cells

2025

This video demonstrates the immunofluorescence staining of a transgenic Drosophila brain for single-cell imaging of glial cells. The brain expresses membrane-targeted proteins fused with different combinations of antigenic epitopes under the control of a glial-specific recombinase system. The isolated and fixed brains are stained with primary antibodies specific to the antigenic epitopes, followed by fluorophore-conjugated secondary antibodies. This labeling enables the simultaneous imaging of...
