Human Astrocytes

Human astrocytes are specialized glial cells in the central nervous system that support neuronal function, maintain tissue homeostasis, and shape brain signaling. Their branched processes contact synapses and blood vessels, where they regulate extracellular ions, remove neurotransmitters such as glutamate, provide metabolic support, and contribute to the blood-brain barrier; activity-dependent calcium signaling enables communication with neighboring cells. Studying human astrocytes helps clarify neural circuit function, synaptic regulation, and responses to injury or disease. Human cell cultures, induced pluripotent stem cell models, and brain organoids also support research into neurodegenerative disorders, neurodevelopmental conditions, and potential therapeutic strategies.

Source: Creighton, B. A., et al. Visualizing and Analyzing Intracellular Transport of Organelles and Other Cargos in Astrocytes. J. Vis. Exp. (2019)This video demonstrates the visualization of intracellular cargo transport in cultured astrocytes using a fluorescent acidotropic probe that selectively labels acidic endolysosomal vesicles. Time-lapse imaging and Z-stack acquisition with a confocal microscope allow tracking of fluorescently labeled cargo throughout the three-dimensional cell volume.
