Minutien Pin Penetration

Minutien pin penetration is a microsurgical technique that uses a fine, pointed pin to create a controlled opening in small biological specimens, providing precise physical access for neuroscience experiments. The operator positions the specimen under magnification and advances the pin through the cuticle or other protective tissue to a defined depth, while minimizing unintended mechanical damage. This approach can support neural tissue access, localized delivery of experimental substances, or reproducible injury models in insects and other small preparations. Its low-cost design and fine spatial control make it useful for studying neural responses, tissue repair, and the effects of traumatic damage.

This video demonstrates the induction of penetrating traumatic brain injury in the drosophila brain. Newly emerged flies are selected, anesthetized, and positioned under a fluorescence-equipped stereo microscope. The GFP-expressing mushroom body is identified, and a Minutien pin is inserted to cause injury. The flies are allowed to recover for further studies.
