Motor Threshold Determination

Motor threshold determination is a neurophysiological method used to identify the minimum stimulation intensity needed to activate the motor cortex and produce a measurable muscle response. In transcranial magnetic stimulation, researchers deliver pulses over the motor cortex while recording motor-evoked potentials with electromyography, then adjust intensity until responses meet a defined consistency criterion during rest or voluntary muscle contraction. This individualized threshold helps standardize stimulation across participants and sessions. It supports studies of corticospinal excitability, motor-system organization, neuroplasticity, and the effects of neurological disease or rehabilitation, while also guiding therapeutic and experimental stimulation protocols.

Here, we describe a rapid reliable and simple procedure to determine the lowest temperature at which rats or mice show nocifensive behavior, i.e. the thermal nociceptive threshold (TNT). This method applies a slowly increasing thermal stimulus allowing precise and reproducible estimation of TNTs with minimum, if any, stress to the animals.

Algorithms assessing heat and mechanical pain thresholds in experimentally inflamed skin of human study subjects are shown. The two pain testing paradigms independently examine nociceptive processing by the two major peripheral nerve fiber populations transmitting pain, i.e., non-myelinated C fibers and small myelinated A-delta fibers.

Human pain models are valuable tools used to assess the analgesic potential of novel compounds and predict their clinical efficacy, especially when used in an integrated manner. Although implementation of these models is complex, with proper execution, the pain models described in this protocol can provide predictive and reliable results.

We propose a method to measure a parameter that is highly relevant for corrosion assessments or predictions of reinforced concrete structures, with the main advantage of permitting testing of samples from engineering structures. This ensures real conditions at the steel-concrete interface, which are crucial to avoid artifacts of laboratory-made samples.
