Two-phase Extraction

Two-phase extraction is a separation method that distributes compounds between two immiscible liquid phases, typically an aqueous phase and an organic solvent, according to their chemical properties. During processing, mixing increases contact between the phases, while differences in polarity, solubility, ionization, and partition coefficients drive target molecules into one phase; centrifugation or settling then enables phase separation. In neuroscience, this approach helps isolate lipids, metabolites, drugs, and other small molecules from brain tissue, cerebrospinal fluid, or cell samples before analytical techniques such as chromatography and mass spectrometry. Careful solvent selection and pH control improve recovery and support reliable biochemical and neurochemical measurements.
