Calcium-sensitive Fluorescent Dye

Calcium-sensitive fluorescent dyes are molecular indicators that report changes in intracellular calcium concentration, a key signal in processes such as muscle contraction, neurotransmitter release, and pharmacological responses. These dyes enter cells and bind Ca2+, causing a measurable change in fluorescence intensity or emission properties; researchers can monitor this signal with fluorescence microscopy, plate readers, or flow cytometry. In pharmacology, calcium-sensitive fluorescent dyes help characterize drug effects on ion channels, receptors, excitation-contraction coupling, and intracellular signaling. Time-resolved calcium measurements can reveal changes in cellular activity, support compound screening, and clarify mechanisms underlying therapeutic or toxic responses.

Assessing T-cell Receptor-Induced Calcium Influx Using a Calcium Indicator Dye

This video analyzes T-cell receptor-induced calcium influx in T-cells. Anti-CD3 antibodies activate the T-cell signaling pathways, leading to an increase in intracellular calcium ions that bind with Indo-1 dye. In flow cytometry, a spectrum shift from the calcium-free state to the calcium-bound state correlates with T-cell receptor-induced calcium influx.

Targeted-esterase induced dye loading (TED) supports the analysis of intracellular calcium store dynamics by fluorescence imaging. The method bases on targeting of a recombinant Carboxylesterase to the endoplasmic reticulum (ER), where it improves the local unmasking of synthetic low-affinity Ca2+ indicator dyes in the ER lumen.

Here, we describe a method for loading a calcium-sensitive dye through the frog nerve stump into the nerve endings. We also present a protocol for the recording and analysis of fast calcium transients in the peripheral nerve endings.
