Long-term Dynamics

Long-term dynamics are patterns of stability, change, and reciprocal influence that unfold over extended periods, making them central to understanding how psychological characteristics and behavior develop. In psychology, researchers examine these dynamics through longitudinal designs, repeatedly measuring individuals or groups to track trajectories and test how experiences, developmental processes, and changing environments interact over time. Such analyses can distinguish temporary fluctuations from enduring change and reveal feedback loops in which behavior affects context while context reshapes behavior. Long-term dynamics inform research on personality, mental health, learning, relationships, and development, supporting more accurate theories, risk assessment, and interventions tailored to when and how psychological change occurs.

Here we present a protocol for live-imaging wound repair and the associated inflammatory response at high spatio-temporal resolution in vivo. This method utilizes the pupal stage of Drosophila development to enable long-term imaging and tracking of specific cell populations over time and is compatible with efficient RNAi-mediated gene inactivation.

We present a step-by-step protocol for the isolation of long-term hematopoietic stem cells (LT-HSCs) and short-term HSCs (ST-HSCs) using the Hoxb5 reporter system.

In Vitro Calcium Imaging in Short-Term and Long-Term Rat Hippocampal Cultures

Source: Calvo-Rodríguez, et al. Fluorescence and Bioluminescence Imaging of Subcellular Ca2+ in Aged Hippocampal Neurons. J. Vis. Exp. (2015). This video demonstrates the use of fluorescence imaging to measure cytosolic calcium levels in short-term and long-term rat hippocampal cultures. By comparing young and aged neurons, it highlights how increased L-type voltage-gated calcium channel density and prolonged activation in aged neurons result in calcium overloading, a hallmark of neuronal aging.

Visualization of Cortex Organization and Dynamics in Microorganisms, using Total Internal Reflection Fluorescence Microscopy

Total Internal Reflection Fluorescence (TIRF) microscopy is a powerful approach to observe structures close to the cell surface at high contrast and temporal resolution. We demonstrate how TIRF can be employed to study protein dynamics at the cortex of cell wall-enclosed bacterial and fungal cells.

C. elegans Positive Butanone Learning, Short-term, and Long-term Associative Memory Assays

Here we describe methods to test C. elegans associative learning and short- and long-term associative memory. These population assays employ the worms abilities to chemotax toward volatile odorants, and form positive associations upon pairing food with the chemoattractant butanone. Increasing the number of conditioning periods induces long-term memory.
