Mechanical Protein Functions

Mechaniczne funkcje białek są napędzane przez konwersję energii chemicznej na pracę mechaniczną poprzez konformacyjne zmiany w strukturze białka.
W domenie białkowej proste zmiany chemiczne, takie jak hydroliza związanych cząsteczek, takich jak ATP lub GTP, mogą powodować zmiany konformacyjne, które powodują znacznie większe ruchy w białku.

Białka pełnią w komórce wiele funkcji mechanicznych. Białka te można podzielić na dwie ogólne kategorie – białka generujące siły mechaniczne i białka poddawane działaniu sił mechanicznych. Białka zapewniające mechaniczne wsparcie strukturze komórki, takie jak keratyna, poddawane są działaniu siły mechanicznej, natomiast białka biorące udział w ruchu komórkowym i transporcie cząsteczek przez błony komórkowe, takie jak pompa jonowa, są przykładami generowania siły mechanicznej. Funkcje takie jak ruch komórek i skurcze mięśni wymagają zmianę energii chemicznej na mechaniczną, zwykle poprzez zmiany konformacyjne. Proteins can create and respond to mechanical force in cells. Some proteins generate force, while others are pulled or stretched by it. Proteins such as keratin provide structural support and are subjected to mechanical force. Proteins such as ion pumps help move molecules across cell membranes and can generate mechanical force.

Cell movement and muscle contraction depend on turning chemical energy into motion. This often happens through conformational change, which is a shift in a protein’s shape. Hydrolysis of nucleoside triphosphates, such as ATP and GTP, can cause a small change that is then amplified into a much larger structural shift.

EF-Tu shows how this works in protein transport. EF-Tu has three distinct domains and transfers a tRNA molecule to the ribosome. One domain binds GTP, and when GTP is hydrolyzed to GDP, the nucleotide-binding site changes because inorganic phosphate is released. This triggers movement of an alpha-helix at the interface of the GTP domain and the other two domains, which changes the positions of the domains and helps release the tRNA so it can enter the ribosome.

Actin also plays several mechanical roles. It can serve as a track for myosin to move along. Depending on the type, myosin may pull on actin filaments or carry an attached organelle along the filament. As part of the cytoskeleton, actin filaments also support cell structure. During cell movement, they push on the cell membrane and help form filopodia and lamellipodia, which are membrane extensions that let the cell move to a new location. Scientists can measure the force actin produces during membrane deformation with tools such as optical tweezers.

Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
