Transgenic Plants

Rośliny transgeniczne są wytwarzane przez wprowadzenie obcego genu do genomu rośliny. Rośliny te mogą być uprawiane z różnych powodów, w tym odporności na wirusy lub szkodniki, wyższych plonów lub produkcji białka terapeutycznego.
Aby wyprodukować roślinę transgeniczną, gen będący przedmiotem zainteresowania i jego promotor są wprowadzane do protoplastu roślinnego, komórki roślinnej bez ściany komórkowej, poprzez inkubację obu w pożywce zawierającej glikol polietylenowy.
Rekombinowany protoplast jest hodowany na sterylnym podłożu w obecności regulatorów wzrostu i składników odżywczych.

Technologia rekombinacji DNA zwana transgenezą jest często stosowana w celu dodania obcego genu lub usunięcia szkodliwego genu z organizmu. Takie organizmy zmodyfikowane nazywane są organizmami transgenicznymi. Pierwszą w historii rośliną transgeniczną była roślina tytoniu opracowana w 1983 r., która wykazała odporność na wirusa mozaiki tytoniowej. Od tego czasu opracowano i wprowadzono na rynek wiele roślin transgenicznych w celu poprawy wartości rolniczej, ozdobnej i ogrodniczej roślin uprawnych. Rośliny transgeniczne można również wykorzystać jako bioreaktor do produkcji ważnych ekonomicznie metabolitów lub białek. <h4>Fizyczne metody przenoszenia genów do roślin</h4> Do przeniesienia obcego genu do protoplastu roślinnego można zastosować wiele metod, takich jak elektroporacja i bombardowanie mikropociskami. Podczas elektroporacji urządzenie dostarcza impulsy wysokiego napięcia do zawiesiny protoplastów. Zwiększa to przepuszczalność protoplastu, umożliwiając obcemu DNA przedostanie się do komórki i integrację z genomem rośliny. Natomiast podczas bombardowania mikropociskami lub metody biolistycznej kuliste cząstki wykonane z wolframu lub złota powlekane są interesującym genem niosącym plazmid. Powleczone cząstki są następnie przyspieszane do 300 do 600 m/s za pomocą instrumentu zwanego działem cząstek. Przyrząd wykorzystuje hel pod wysokim ciśnieniem, aby zapewnić siłę napędową, która pomaga cząstkom pokrytym plazmidem przeniknąć przez ścianę komórkową rośliny. Po wejściu do komórki interesujący gen integruje się z genomem rośliny. Metodę tę można zastosować do wprowadzenia DNA do hodowli kalusa, tkanek merystematycznych, niedojrzałych zarodków, zawiesin komórek roślinnych itp. <h4>Zastosowania roślin transgenicznych</h4> Technologia rekombinacji DNA jest szeroko stosowana w opracowywaniu roślin odpornych na szkodniki i wirusy. Technika ta jest również pomocna w modyfikowaniu zawartości lipidów, skrobi i białka w nasionach. Na przykład złoty ryż to genetycznie zmodyfikowana roślina ryżu o zwiększonej wartości odżywczej. Ta transgeniczna odmiana ryżu wytwarza β-karoten – prekursor witaminy A, nadający mu złocisty kolor. Jest wytwarzany w wyniku transformacji DNA za pośrednictwem Agrobacterium, podczas której rekombinowany DNA niosący trzy geny kodujące enzymy – syntazę fitoenu, desaturazę fitoenu i β-cyklazę likopenu, wprowadza się do genomu docelowych zarodków ryżu. Zarodki następnie rozwijają się w rośliny wytwarzające β-karoten. Złoty ryż został opracowany w celu uzupełnienia niedoborów witaminy A w krajach rozwijających się. Niektóre inne rośliny transgeniczne, takie jak bawełna Bt, zostały zmodyfikowane poprzez wstawienie genu wyizolowanego z bakterii Bacillus thuringiensis – bakterii Gram-dodatniej występującej w glebie. Takie rośliny transgeniczne wytwarzają białko krystaliczne, czyli białko „Cry”, które zabija szkodliwe owady, czyniąc roślinę odporną na owady.

Transgenic plants are crops that have been changed with recombinant DNA technology, also called transgenesis. This method adds a foreign gene or removes a harmful one from an organism. The result is a genetically modified organism. The first transgenic plant was a tobacco plant made in 1983, and it was resistant to tobacco mosaic virus.

Since then, transgenic plants have been developed for many uses in agriculture, horticulture, and ornamental plants. Some are designed to resist pests or viruses. Others are used to improve the lipid, starch, or protein content of seeds. In some cases, transgenic plants can also act as bioreactors and make useful metabolites or proteins.

Foreign genes can be transferred into plant protoplasts by physical methods such as electroporation and microprojectile bombardment. In electroporation, high-voltage pulses make the protoplast membrane more permeable. This allows the foreign DNA to enter the cell and integrate into the plant genome. In microprojectile bombardment, also called the biolistic method, tungsten or gold particles are coated with plasmid DNA and shot into the cells with a particle gun.

The particle gun uses high-pressure helium gas to accelerate the coated microprojectiles to about 300 to 600 m/s. The particles can penetrate the plant cell wall and deliver the gene of interest. This method can be used with callus cultures, meristematic tissues, immature embryos, and plant cell suspensions. Once inside the cell, the introduced DNA can integrate into the plant genome.

A well-known example is golden rice, which has enhanced nutritional value. It produces β-carotene, a precursor of vitamin A, and this gives the rice its golden color. Golden rice was made by Agrobacterium-mediated DNA transformation using recombinant DNA with genes for phytoene synthase, phytoene desaturase, and lycopene β-cyclase. It was developed to help address vitamin A deficiency in developing nations.

Another example is Bt cotton. It contains a gene from Bacillus thuringiensis, a gram-positive soil bacterium. The plant makes Cry protein, which kills harmful insects and makes the crop insect resistant. These examples show how transgenic plants can improve crop value and help solve real agricultural problems.
