The Phragmoplast

Komórki roślinne są otoczone sztywną ścianą komórkową, co utrudnia komórkom podział poprzez kurczenie się z zewnątrz. W związku z tym komórki roślinne dzielą się, tworząc nową ścianę komórkową między dwoma jądrami potomnymi w sposób "na lewą stronę". Wyspecjalizowana struktura roślin zwana fragmoplastem kieruje tworzeniem nowej ściany komórkowej.
Płaszczyzna podziału komórki, która jest przyszłym miejscem tworzenia się ściany komórkowej, jest oznaczona pasmem preprofazy składającym się z mikrotubul i włókien aktynowych, które tworzy się podczas fazy G1 cyklu komórkowego. Pasma preprofazowe gromadzi się w korze komórkowej i znika na początku fazy M.
Podczas mitozy chromosomy rozdzielają się i zaczynają przemieszczać się na przeciwległe bieguny. Na biegunach wokół oddzielonych chromosomów tworzy się otoczka jądrowa, a wrzeciona zaczynają zanikać. Pozostałe mikrotubule wrzeciona mitotycznego tworzą fragmoplast.
Mikrotubule fragmoplastów są biegunowe, z końcami dodatnimi w pobliżu płaszczyzny równikowej i końcami ujemnymi w pobliżu biegunów.
Ciała Golgiego wydzielają małe pęcherzyki, które są przenoszone przez białka motoryczne wzdłuż mikrotubul do centrum komórki. Pęcherzyki te są wypełnione polisacharydami i glikoproteinami, dwoma ważnymi składnikami wymaganymi do syntezy nowej ściany komórkowej. Fuzja pęcherzyków tworzy strukturę przypominającą dysk zwaną wczesną płytką komórkową.
Mikrotubule fragmoplastów są z powodzeniem regenerowane na wolnych brzegach płytki komórkowej, umożliwiając fragmoplastowi boczne wydłużenie płytki. Podział komórek jest niezbędny do wzrostu i rozwoju organizmu. W komórkach zwierzęcych wrzeciono centralne i związane z nim białka tworzą ciało środkowe, strukturę odgrywającą zasadniczą rolę w cytokinezie. U roślin wrzeciono centralne wraz z mikrotubulami, aktyną i innymi składnikami komórek dojrzewa do fragmoplastu, który jest niezbędny do cytokinezy. W przeciwieństwie do stacjonarnego korpusu środkowego, fragmoplast rozszerza się odśrodkowo, co ostatecznie prowadzi do powstania nowej ściany komórkowej. Dojrzały fragmoplast ma kształt pączka i zawiera zewnętrzny obszar wiodący, środkową strefę przejściową i wewnętrzny obszar opóźniony. W regionie wiodącym nowe mikrotubule są montowane w sposób ciągły. Zarodkowanie mikrotubul inicjowane jest przez kompleks pierścieniowy γ-tubuliny (γ-TuRC), który zawiera sześć podjednostek – GCP1 do GCP6. Jednakże γ-TuRC nie może bezpośrednio oddziaływać z mikrotubulami; dlatego augmin, kompleks białkowy oddziałujący z mikrotubulami, pośredniczy w rekrutacji γ-TuRC do mikrotubul. Mikrotubule obecne w fragmoplastach pełnią rolę torów transportu pęcherzyków wydzielniczych wytwarzanych przez pobliskie ciało Golgiego. W środkowej strefie przejściowej pęcherzyki początkowo przekształcają się w struktury kanalikowe, a następnie łączą się, tworząc splecioną sieć rurową. Ciągłe dodawanie pęcherzyków wydzielniczych powoduje wzrost sieci kanalików na zewnątrz, aż do zetknięcia się z błoną komórki macierzystej. Błony pęcherzyków służą do tworzenia błony plazmatycznej komórek potomnych, podczas gdy biomolekuły obecne w pęcherzykach służą do budowy płytki komórkowej znajdującej się pomiędzy błonami dwóch komórek potomnych. W obszarze opóźnionym mikrotubule ulegają depolimeryzacji, pozostawiając dojrzałą część płytki komórkowej. W pełni uformowana płytka komórkowa rozwija się następnie w ścianę komórkową oddzielającą dwie komórki potomne. Polimeryzacja mikrotubul na przednim końcu i depolimeryzacja mikrotubul w obszarze opóźnionym powodują ekspansję na zewnątrz fragmoplastu i płytki komórkowej, aż ta ostatnia połączy się z macierzystą ścianą komórkową. Nieprawidłowe funkcjonowanie fragmoplastów może skutkować powstaniem komórek wielojądrzastych i fenotypów śmiertelnych dla embrionów.

Plant cells build a new wall during cytokinesis with help from the phragmoplast. This plant structure forms from the central spindle and related cell parts as the cell finishes dividing. In animal cells, the comparable structure is the midbody, but in plants the phragmoplast expands outward to guide new wall formation.

The mature phragmoplast has a donut-like shape. It has an outer leading region, a middle transition zone, and an inner lagging region. New microtubules are assembled continuously in the leading region. Microtubule nucleation begins with the gamma-tubulin ring complex, or gamma-TuRC, which contains six subunits called GCP1 through GCP6. Because gamma-TuRC cannot attach to microtubules on its own, augmin, a microtubule-interacting protein complex, helps recruit it to the microtubules.

The microtubules in the phragmoplast act as tracks for secretory vesicles from the nearby Golgi body. In the middle transition zone, these vesicles first change into tubule-like structures. They then connect to form an intertwined tubular network. As more vesicles are added, the network grows outward until it reaches the parental cell membrane.

The vesicle membranes become the plasma membrane of the daughter cells. The materials inside the vesicles help build the cell plate between the two daughter-cell membranes. In the lagging region, the microtubules break down, leaving the mature part of the cell plate behind. The finished cell plate then develops into the cell wall that separates the two daughter cells.

This outward growth depends on microtubules being built at the leading end and broken down at the lagging end. If phragmoplast function fails, cells can become multinucleated. It can also lead to embryonic lethal phenotypes.
