Glucose Depletion

Glucose depletion is a biochemical state in which available glucose falls below the level required to sustain cellular energy production and biosynthetic activity. As glucose becomes scarce, cells reduce glycolytic flux and may mobilize glycogen or increase gluconeogenesis, while shifting toward alternative carbon sources and activating energy-conservation pathways. Studying glucose depletion helps clarify metabolic regulation, nutrient sensing, and cellular adaptation to changing energy supplies. In biochemistry and related research, controlled glucose withdrawal is used to investigate pathway responses, compare cellular phenotypes, and assess how altered glucose availability affects growth, survival, and macromolecule synthesis.

In this video, we demonstrate an ex vivo technique to measure extracellular glucose analog depletion in harvested mouse organs. It involves incubating the mouse tissue in a solution containing fluorescently-labeled deoxyglucose, which is taken up by the cells. The glucose analog uptake can be indirectly determined by measuring the fluorescence of the incubated medium.
