Hydrogel Formulation

Hydrogel formulation is the design and preparation of water-rich, three-dimensional polymer networks that mimic key features of soft biological tissues and support biomedical engineering applications. It typically combines hydrophilic polymers with a crosslinking process, such as covalent bonding, ionic interactions, or physical assembly, to control swelling, mechanical strength, porosity, degradation, and molecular transport. By adjusting polymer concentration, crosslinker chemistry, and processing conditions, researchers can create materials that encapsulate cells, proteins, or therapeutic compounds while maintaining a suitable aqueous environment. These formulations are used in tissue engineering, drug delivery, wound care, and three-dimensional cell culture, where their tunable properties influence biocompatibility, release behavior, and tissue integration.

Supramolecular hydrogelators based on ureido-pyrimidinones allow full control over the macroscopic gel properties and the sol–gel switching behavior using pH. Here, we present a protocol for formulating and injecting such a supramolecular hydrogelator via a catheter delivery system for local delivery directly in relevant areas in the pig heart.

Freeze-drying is often an easy and convenient way to obtain dry products of viable bacterial cells. An issue of the process is cell survival. We detail here a procedure to investigate how cell survival during freeze-drying is influenced by the properties of the formulation used.
