In Vitro Bone

In vitro bone refers to bone tissue or bone-like constructs developed and maintained outside a living organism, providing a controllable model for studying skeletal biology and engineering. Researchers typically culture osteogenic cells on biomaterial scaffolds or within three-dimensional matrices, where biochemical signals and mechanical conditions promote extracellular matrix production, collagen organization, and mineralization. These models support investigation of bone formation, remodeling, disease mechanisms, and interactions between cells and biomaterials. In bioengineering, in vitro bone systems also enable evaluation of regenerative strategies, implant materials, and therapeutics under controlled laboratory conditions, potentially reducing reliance on animal studies and improving the design of patient-relevant bone substitutes.

This video demonstrates the differentiation of bone marrow stromal cells (BMSCs) into adipocytes. In the two-step process, in the presence of specific growth factors and hormones, undifferentiated BMSCs first differentiate into preadipocytes. The second step involving treatment with glucose and insulin leads to the formation of mature adipocytes with multiple lipid droplets.
