Mechanical Analysis

Mechanical analysis is the quantitative study of how materials, structures, and biological systems respond to forces, motion, and deformation. It uses measurements such as stress, strain, stiffness, elasticity, and viscoelasticity to relate applied loads to changes in shape or function, often through controlled experiments and mechanical models. In bioengineering, these analyses characterize tissues, cells, biomaterials, implants, and engineered constructs under conditions that approximate their physiological environment. The resulting data guide the design of medical devices and tissue scaffolds, improve understanding of structure-function relationships, and help evaluate safety, durability, and compatibility.

Myo-mechanical Analysis of Isolated Skeletal Muscle

To assess the in vivo effects of therapeutic interventions for muscle disease, methods are needed to quantitate force generation and fatigability in treated muscle. We detail an approach to evaluating myo-mechanical properties in explanted mouse hindlimb muscle. This analysis provides a robust approach to quantitating the effects of genetic modification on muscle function, as well as comparison of therapies in mouse models of muscle disease.

Mechanical Stimulation of Chondrocyte-agarose Hydrogels

The biosynthesis of cartilaginous extracellular matrix by chondrocytes can be affected by application of mechanical stimuli. This method describes the technique of applying dynamic compressive strains to chondrocytes encapsulated in 3D constructs and the evaluation of induced changes in chondrocyte metabolism.

Mechanical Dissociation of Tissues for Single Cell Analysis Using a Motorized Device

A general protocol for the combined enzymatic and semi-automated mechanical dissociation of tissues to generate single-cell suspensions for downstream analyses, such as flow cytometry, is provided. Instructions for the fabrication, assembly, and operation of the low-cost mechanical device developed for this protocol are included.

Chemical reactions often occur in a stepwise fashion, involving two or more distinct reactions taking place in a sequence. A balanced equation indicates the reacting species and the product species, but it reveals no details about how the reaction occurs at the molecular level. The reaction mechanism (or reaction path) provides details regarding the precise, step-by-step process by which a reaction occurs. In this protocol, we demonstrate the fabrication of a microactuator array of vertically displaced posts on which the technology is based, and how this base technology can be modified to conduct high-throughput mechanically dynamic cell culture in both two-dimensional and three-dimensional culture paradigms.
