Real-time Cell Manipulation

Real-time cell manipulation is the observation and controlled handling of living cells while their behavior is monitored, enabling researchers to respond to changes as they occur. In bioengineering, integrated imaging, microfluidic systems, and tools such as optical, electrical, or mechanical actuators can position cells, alter their local environment, or sort them based on measured properties, with feedback linking detection to intervention. This approach supports precise studies of cell signaling, migration, viability, and interactions with engineered materials, while reducing reliance on fixed, end-point measurements. It can improve single-cell analysis, cell therapy development, tissue engineering, and the design of responsive biological systems.

A procedure is described for manipulating the activity of cerebral cortical pyramidal neurons optogenetically while the electroencephalogram, electromyogram, and cerebral lactate concentration are monitored. Experimental recordings are performed on cable-tethered mice while they undergo spontaneous sleep/wake cycles. Optogenetic equipment is assembled in our laboratory; recording equipment is commercially available.
