Rotating Wall Vessel

A rotating wall vessel is a bioreactor that suspends cells, tissues, or engineered constructs in a continuously rotating, fluid-filled vessel, creating a low-shear environment for three-dimensional culture. Rotation keeps the culture contents in free fall relative to the vessel wall, reducing sedimentation and limiting damaging mechanical forces while maintaining nutrient and gas exchange. In bioengineering, this system supports the formation of tissue-like aggregates, organoids, and other three-dimensional models that more closely replicate aspects of native tissue architecture than conventional monolayer culture. Rotating wall vessels are used to study cell behavior, tissue development, disease processes, and the performance of biomaterials under controlled laboratory conditions.

A rotating cell culture system that allows epithelial cells to grow under physiological conditions resulting in 3-D cellular aggregate formation is described. The aggregates generated display in vivo-like characteristics not observed in conventional culture models and serve as a more accurate organotypic model system for a multitude of scientific investigations.
