Stem Cell Recruitment

Stem cell recruitment is the directed attraction and movement of stem cells toward a target tissue, where they can support repair, regeneration, or functional recovery. The process depends on signaling gradients created by injured or engineered tissues, including chemokines, growth factors, extracellular matrix cues, and cell-adhesion interactions that guide stem-cell migration and retention. In bioengineering, researchers incorporate these signals into biomaterial scaffolds, tissue-engineered constructs, and therapeutic delivery systems to recruit endogenous stem cells to damaged sites. Effective recruitment may improve tissue integration, reduce reliance on transplanted cells, and advance regenerative strategies for repairing complex tissues.

Analyzing the Effects of Stromal Cells on the Recruitment of Leukocytes from Flow

The ability of inflamed endothelium to recruit leukocytes from flow is regulated by mesenchymal stromal cells. We describe two in vitro models incorporating primary human cells that can be used to assess neutrophil recruitment from flow and examine the role that mesenchymal stromal cells play in regulating this process.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

In a parallel plate flow chamber model, pro-inflammatory cytokines induce the expression of adhesion receptors by endothelial cells, facilitating neutrophil recruitment and migration. The cross-talk between stromal cells and endothelial cells diminishes neutrophil recruitment to the endothelial cells.
