Gfp Tagged Ribosomal Subunit

A GFP-tagged ribosomal subunit is a ribosome component genetically fused to green fluorescent protein (GFP), enabling researchers to visualize and track ribosomes in living cells. The GFP fusion produces fluorescence when excited with appropriate light, while the labeled ribosomal protein remains associated with its subunit and can report its localization, movement, assembly, or exchange under experimental conditions. In biology, this approach supports studies of protein synthesis, ribosome distribution, and cellular responses to environmental or physiological changes. Here, we describe a detailed protocol for the in vitro reassociation of purified, salt-washed eukaryotic ribosomal subunits for the analysis of 80S particle formation. This method is illustrated by analyzing ribosomal subunits isolated from a wild-type strain of Saccharomyces cerevisiae and a mutant strain deficient in the ribosomal protein eL24.
