Egfp Tagging

EGFP tagging is a molecular biology technique that links the gene for enhanced green fluorescent protein (EGFP) to a gene of interest, allowing the resulting protein to be visualized in living cells or tissues. After expression, the target protein and EGFP are produced as a fusion; EGFP folds into a fluorescent chromophore that emits green light when excited, so fluorescence microscopy can reveal the protein’s location, movement, and relative distribution. In biology, this approach supports studies of protein trafficking, cell organization, and dynamic cellular processes, while helping researchers assess how genetic changes affect protein behavior. Careful tag placement and expression control help preserve normal function.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.
