Lens Transparency

Lens transparency is the ability of the eye’s crystalline lens to transmit and focus light with minimal scattering, a property essential for clear vision. It depends on densely packed, precisely organized crystallin proteins, the removal of light-scattering organelles from mature lens fiber cells, and maintenance of a stable refractive index across the tissue. Disruption of protein organization, hydration, or cellular maintenance can increase light scattering and produce cataracts, an important cause of visual impairment. Studying lens transparency helps explain eye development, aging, and cataract formation while informing research on lens physiology, disease mechanisms, and strategies for preserving or restoring vision.

We present an efficient method of studying lens accommodation by using a manual lens stretcher. The protocol mimics physiological accommodation by pulling the zonules connected around the lens capsule, thereby, stretching the lens.

RNA Isolation from Mouse Ocular Lens Epithelium and Fiber Cell Bulk Masses

2025

This protocol details the separation of the mouse ocular lens epithelium and fiber cell bulk mass, followed by RNA isolation and qPCR analysis. This method allows for separating lens cell compartments for a more detailed analysis of the transcriptome and biological processes in epithelial cells vs. differentiated fiber cells.
