Acidic Organelle Compartment

An acidic organelle compartment is a membrane-bound region of a cell that maintains a lower pH than the surrounding cytoplasm, enabling specialized chemical reactions and cargo processing. Proton-pumping V-type ATPases transport hydrogen ions across the organelle membrane, while selective membrane permeability and ion transport help sustain the acidic environment. Endosomes, lysosomes, and plant vacuoles use this condition to sort and degrade proteins, activate hydrolytic enzymes, and regulate the movement of materials through the endomembrane system. Studying acidic organelle compartments clarifies intracellular trafficking, nutrient recycling, membrane turnover, and mechanisms underlying diseases linked to defective lysosomal or endosomal function.

Herein we describe two approaches to characterize cell polarization events in B lymphocytes during the formation of an IS. The first, involves quantification of organelle recruitment and cytoskeleton rearrangements at the synaptic membrane. The second is a biochemical approach, to characterize changes in composition of the centrosome, which undergoes polarization to the immune synapse.

Several methods are available for measuring intracellular pH but very few of these allow simultaneous measurement of cytoplasmic and organellar pH. Here, we describe in detail a rapid and accurate methodology to simultaneously measure cytoplasmic and vesicular pH by ratiometric imaging of living cells.
