Murine Hindlimb

The murine hindlimb is the hind leg of a mouse, a compact anatomical system used to study movement, musculoskeletal organization, and tissue function in mammals. Its bones, joints, skeletal muscles, nerves, and blood vessels work together as muscle contractions generate force across the hip, knee, and ankle to produce locomotion and maintain posture. In biology, murine hindlimb models support research on limb development, neuromuscular coordination, bone and muscle injury, vascular function, and regeneration. Structural analysis, gait assessment, and experimental manipulation of the hindlimb can reveal how tissues develop, respond to disease or injury, and recover after treatment.

Methods for Acute and Subacute Murine Hindlimb Ischemia

Surgical induction of hindlimb ischemia in the mouse is useful to examine angiogenesis, however this is compromised in certain inbred mouse strains that display marked ischemia-induced tissue necrosis. Methods are described to induce subacute limb ischemia using ameroid constrictors to circumvent this problem through the induction of gradual arterial occlusion.

We present a reproducible mouse model of heterotopic hindlimb-to-neck vascularized composite allotransplantation. This supermicrosurgical protocol, involving 0.3-0.5 mm vessel anastomoses, integrates nylon filament guidance and venous cuffing. It provides a reliable platform for translational studies on tolerance, ischemia-reperfusion injury, and immunological mechanisms in vascularized composite allotransplantation.
