Offspring Cup Labeling

Offspring cup labeling is a biological record-keeping method used to identify and track newly produced offspring and the conditions associated with them. Researchers assign each cup a clear, unique label, typically linking its contents to a parental pair, collection date, experimental group, or treatment, and maintain consistent records throughout handling and observation. Accurate labeling helps prevent sample mix-ups, preserves information about lineage and rearing conditions, and supports reliable comparisons of survival, development, behavior, or other measured traits. In breeding studies and laboratory experiments, it strengthens data quality and makes results easier to reproduce and interpret.

This protocol describes an approach for in toto labeling and multidimensional imaging of zebrafish early eye development. We describe labeling, embedding, and four dimensional (4D) imaging using laser scanning confocal microscopy, and considerations for optimizing acquisition of datasets for dissecting mechanisms of optic cup morphogenesis.

We discussed the synthesis of individual graphitic nanocups using a series of techniques including chemical vapor deposition, acid oxidation and probe-tip sonication. By citrate reduction of HAuCl4, the graphitic nanocups were effectively corked with gold nanoparticles due to the chemically reactive edges of the cups.

This protocol presents a step-by-step guide for the Simultaneous Label-free Autofluorescence Multi-harmonic (SLAM) microscopic technique, including details on how to generate the laser light source, prepare a tissue sample, conduct imaging, and analyze the data. SLAM advances nonlinear microscopy by measuring four complementary label-free contrasts to investigate the tissue microenvironment.
